Scientist in Protein Technologies Center - Cell Engineering / Molecular Biology

The Protein Technologies Center (PTC) within the Department of Structural Biology at St. Jude Children’s Research Hospital seeks a Scientist with expertise in saturation‑mutagenesis functional assays, site‑specific integrase‑based stable expression platforms, and genome‑editing technologies. Responsibilities Engineer cell lines using CRISPR gene‑editing technology for generating gene knockouts and knock‑ins (SNVs, gene tags, etc.) and confirm the edits. Generate stable cell lines using non‑viral and viral‑based delivery systems. Implement advanced technologies to develop stable target‑overexpressed cell lines, pools, clones, and NGS library preparation. Perform molecular cloning (In‑Fusion, LIC, PIPE, Gateway, FX, overlap‑extension PCR, Gibson assembly, BioBrick, recombineering) and handle large or complex DNA constructs, including combinatorial libraries, site‑directed and saturation mutagenesis. Validate genomic stability throughout clonal line‑generation and provide pivotal, hands‑on support to structural cell biology projects across the department. Qualifications Minimum education: Bachelor’s degree in a relevant scientific area. Minimum experience: 8years with a Bachelor’s; 6years with a Master’s; 3years with a PhD. Preferred: PhD (with at least 5years of post‑doctoral experience) in molecular biology, synthetic biology, or a related discipline. Proven track record in molecular cloning, large DNA construct handling, combinatorial libraries, site‑directed and saturation mutagenesis.
