SUJET : FRAGMENTATION DU SOMMEIL D’UN MODELE MURIN : IMPACT SUR LE PERMEABILITE DE LA BARRIERE HEMATO ENCEPHALIQUE (BHE)
La fragmentation du sommeil est une composante majeure d’altération des fonctions ‘réparatrices du sommeil’. Elle se retrouve dans des pathologies respiratoires (apnées) ou neurologiques et participe au risque vasculaire cérébral. Au laboratoire, nous avons étudié l’effet d’un stress hypoxique intermittent chronique représentatif de l’apnée du sommeil. Un acteur clé de cette altération est la barrière hémato-encéphalique, barrière qui sépare le compartiment sanguin de cérébral.
La fragmentation du sommeil est donc une composante importante dans l’apnée du sommeil au même titre que l’hypoxie intermittente, mais peu de données sont actuellement disponibles et notamment sur son impact sur la BHE, c’est l’objectif de ce travail de thèse.
L’objectif de ce travail de thèse est une évaluation, sur un modèle de fragmentation du sommeil réalisée sur un modèle murin, de l’impact cérébral anatomique et comportemental. Un monitoring encéphalographique et autonomique cardiaque ,des tests cognitifs, ainsi qu’une analyse des tissus cérébraux sera réalisée afin de quantifier l’effet de cette fragmentation, en particulier l’inflammation systémique consécutive. Une cinétique de récupération ‘post fragmentation’ et des contre-mesures pharmacologiques seront secondairement recherchées.
Le candidat devra maitriser les techniques d’analyses biochimiques et immunohistochimiques classiques profil biosciences. Ce point est un élément de choix qui sera utilisé par le jury. Des compétences et connaissances en neurosciences seraient appréciées. La validation préalable d’une formation réglementaire de niveau 1 et 2 sera apprécié mais non obligatoire. Un travail pratique de M2 sur un modèle murin sera un point d’attention particulière. Le candidat devra faire preuve d’un bon esprit d’équipe et d’une grande curiosité scientifique. Bonne maitrise de l’anglais indispensable.
